Advantages of the Hielscher Microplate Sonicator UIP550MTP

The multi-well plate sonicators UIP250MTP, UIP400MTP and UIP550MTP allow for high-throughput sample sonication in any standard 96-well or ELISA plate.High-Power, Focused Sonication: Achieving consistent and efficient sample disruption in multiwell plates can be difficult when using traditional methods like plate shakers or bead mills. These techniques often produce inconsistent lysate quality, which can negatively impact downstream applications. The Hielscher microplate sonicator UIP550MTP provides a solution by using high-intensity ultrasound waves to create cavitation, ensuring uniform disruption across all wells and resulting in reliable lysate preparation throughout the plate.

High-Throughput: Scalability is another important factor in multiwell plate sample processing. Traditional equipment is often limited in handling multiple plates, making large-scale processing cumbersome. The UIP550MTP sonicator solves this issue by being compatible with standard plate formats (12-, 24-, 48-, 96-, or 384-wells). Its compact design allows for the easy addition of multiple units, enabling high-throughput sample preparation and simultaneous sonication of multiple plates.

Sterile Sonication: Cross-contamination is a frequent concern with traditional methods, as sample splashing or carryover between wells can compromise data quality. The UIP550MTP minimizes this risk by indirectly sonicating the plates, removing the need for beads or chemicals. By covering the plates with foil or a lid, a closed system is created, preventing cross-contamination.

Cost-Efficiency: Cost and flexibility are also key considerations. Many high-throughput sonicators require expensive proprietary plates, which limits options and increases costs. The UIP550MTP, however, works with any standard multiwell plate, allowing researchers to choose the most suitable and cost-effective plates for their experiments, reducing expenses on consumables.

Streamlined Workflows: Traditional sample preparation techniques can be labor-intensive, requiring repetitive manual steps and long processing times. The UIP550MTP sonicator simplifies the workflow, reducing hands-on time and labor. It provides fast, simultaneous treatment of all wells, eliminating the need for multiple incubation steps or manual transfers. Specific sonication protocols can be saved and easily repeated, ensuring consistent and reproducible results.

No Sample Loss: Sample recovery is often challenging with bead mills, as sample loss can occur during bead transfer. The UIP550MTP sonicator offers superior sample recovery and handling by eliminating the need for beads, reducing the complexity of separation steps and minimizing the risk of sample loss or contamination.

Use Different Sample Vessels: The versatility of the UIP550MTP sonicator is another major advantage. It accommodates various plate formats and offers flexible sonication parameters, making it suitable for a wide range of sample types and applications. In addition to multiwell plates, it can be used for small vials, petri dishes, and other containers, for processes such as cell lysis, DNA fragmentation, emulsification, and degassing.

Automation: Finally, the UIP550MTP sonicator integrates seamlessly with automated systems, allowing for smooth incorporation into auto-sampler lines. This compatibility ensures standardized, reproducible sample preparation, boosting efficiency and productivity in high-throughput environments.

Precise Temperature Control During Microtiter Plate Sonication

The UIP550MTP is a powerful 550-watt ultrasonicator that transmits ultrasound waves and vibrations through the walls of a microtiter plate, indirectly affecting the samples. This process creates cavitation and shear forces within the medium. Like other mechanical cell disruption methods, sonication generates heat. However, unlike other techniques, the UIP550MTP offers advanced temperature monitoring and allows users to set temperature limits to prevent thermal degradation of biological samples.
The UIP550MTP is equipped with smart software that can be easily accessed through a digital touchscreen. This interface allows you to pre-set temperature limits and program the total energy input for your sample processing.

Monitoring Sample Temperature: The UIP550MTP features an intelligent software system and a pluggable temperature sensor. By connecting the sensor to the UIP550MTP and placing the sensor tip into one of the microtiter wells, you can specify a temperature range for your sonication process using the digital touchscreen menu. The device will automatically stop when the maximum temperature is reached and pause until the sample cools down to the lower end of the specified temperature range. At that point, sonication will resume automatically. This feature helps prevent heat-induced sample degradation.

To further delay reaching the critical temperature limit, you can pre-cool the 96-well plate, microtiter plate, or ELISA plate and its samples.

What are microtiter plates?

Microtiter plates, also known as 96-well plates, 96-well microplates, multi-well plates or ELISA plates, are rectangular multi-well plates, which are widely used in laboratories to process cultured cells, cellular samples. Microtiter plates are commonly used in manifold assays such as ELISA and PCR. Most 96-well plates are designed for high-throuput workflows and are compatible with analytical instruments and robotic handling and processing. Therefore multiple variants such as fully skirted plates suitable for robotic platforms, non-skirted, semi-skirted, and bar-coded plates adapted to mass sample processing are available.
Common use of microtiter plates include applications such as sample collection, compound preparation, combinatorial chemistry, high throughput screening, nucleic acid purification, bacterial culture growth, and plate replication.

What are ELISA plates?

ELISA plates are flat, multi-well plates typically made of polystyrene, designed for use in enzyme-linked immunosorbent assays (ELISA). These plates allow for the detection and quantification of proteins, antibodies, hormones, or other molecules by immobilizing the target antigen or antibody onto the plate surface. Through a series of binding, washing, and enzyme-substrate reactions, the presence and concentration of the target substance can be measured, often via colorimetric, fluorescent, or luminescent signals.

Can sonication damage cells?

Yes, sonication can damage cells, but the effect of cell disruption depends on the ultrasonic intensity. High-intensity ultrasound waves create cavitation and shear forces that can disrupt cell membranes. While this is beneficial for processes like cell lysis and extraction of intracellular materials, excessive sonication or improper temperature control can lead to unintended cell damage or degradation of sensitive biological samples. However, the Hielscher UIP550MTP multi-well plate sonicator is equipped with intelligent temperature monitoring and control features to minimize heat-induced damage and help ensure more controlled, reproducible cell disruption.
